摘要:

Single-molecule fluorescence spectroscopy has made important application on protein folding, but its time resolution is limited to millisecond. By introducing a photo-protection strategy to protect fluorophores from photobleaching and blinking, we developed a fast single-molecule Foster Resonance Energy Transfer (FRET) approach. Using this method, we resolved sub-milisecond conformational dynamics of two-state protein α-spectrin SH3 and one-state downhill protein BBL that were not accessible with the common single-molecule fluorescence technique. Our experimental results and analysis with Szabo’s photon statistics theory demonstrated the one-state downhill folding scenario of BBL that has been in severe controversy for a decade. Our method also measured the relaxation time of protein conformational fluctuation.
