摘要:

Across phylogeny and ontogeny, pronounced differences are evident in the ability of organisms to regenerate damaged tissues and organs. For example, while lower vertebrates and neonatal mammals retain incredible capacities for heart regeneration, adult mammals generally have limited ability to restore lost cardiac tissue. However, the underlying mechanisms driving loss of such a seemingly beneficial trait in evolution and development have remained enigmatic. Mammalian hearts lose regenerative potential due to cardiomyocyte cell-cycle withdrawal and polyploidization coinciding with the development of postnatal endothermy. Through phylogenetic analysis of vertebrate cardiomyocyte ploidy as a proxy for cardiac regenerative potential, we reveal that diploid cardiomyocyte abundance seems to be constraint by the Kleiber’s law, and decreases when standard metabolic rate, body temperature, and thyroid hormone level increase during the ectotherm-to-endotherm transition. In a parallel targeted chemical screen, we identify thyroid hormones as potent developmental triggers of mammalian cardiomyocyte cell-cycle arrestin vivo. Further pharmacological and genetic manipulations in mice and zebrafish show thyroid hormones inhibit cardiomyocyte proliferation and heart regeneration. Notably, inhibition of thyroid hormone synthesis in mice at birth result in low (31.7°C) body temperatures, resembling that of primitive mammals. Thus, our findings support a model where loss of cardiomyocyte regenerative capacity, driven by increasing thyroid hormone levels, may be a tradeoff for the acquisition of endothermy during animal evolution and postnatal development.

黄果,加州大学旧金山分校生理学系心血管研究所助理教授。 Huang的研究围绕使用心脏作为模型系统和研究成年斑马鱼,新生儿和成年小鼠的器官再生和修复的切入点,重点介绍了调节驻地干细胞活化和成熟细胞去分化/增殖的途径,以及在基因组操作技术,组织和分子工程,单细胞分析和高级成像显微镜方面具有创新和综合的方法。他是心血管研究所的研究员和EliEdythe Broad再生医学和干细胞研究中心的研究员。Huang博士于1999年毕业于北京大学,并在范德比尔特大学学习。他先在约翰霍普金斯大学医学院的生物化学专业获得博士学位然后在德克萨斯大学西南医学中心从事博士后研究。他的研究主要集中在研究可能导致心脏再生潜能的保存的罕见心脏疾病中的人类基因突变和潜在的分子机制。他的研究还侧重于利用phylogenetic screen力量以发现具有非凡组织再生能力的新型高等脊椎动物模型。迄今为止,他在顶尖期刊上发表了多篇杰出的着作,包括Science ,Nature (Research Article),Dev Cell,J Vis Exp,Neuron,Mol Cell,Nature Cell Biology等。
